This pack contains three components:

Topic Mastery Sheet (PDF) - a comprehensive reference covering the effect of temperature, pH and substrate concentration on enzyme activity. Each variable includes an annotated graph with key features labelled: the optimum temperature (~37°C for human enzymes), optimum pH values for amylase (pH 7), pepsin (pH 2) and trypsin (pH 8), and the substrate concentration plateau caused by active site saturation. Investigation methods are described for each variable, including the amylase/starch iodine test practical for pH, the biuret test method for investigating pepsin activity over time, and the hydrogen peroxide/catalase practical for substrate concentration. The rate of reaction formula (Rate = change ÷ time) is covered with a fully worked example: pepsin at 10°C digests 50 g of protein in 36 minutes, giving a rate of 1.4 g/min.

Student Exam Practice Sheet (PDF + DOCX) - five exam-style questions covering all three variables, a rate calculation, interpretation of the biuret test method applied to pepsin at 60°C, and a question on pepsin’s optimum pH and the reason for it.

Teacher Answer Key (PDF + DOCX) - full mark scheme. The 1.4 g/min answer is given with the full working (50 ÷ 36), and a note that rate answers without units will not score the final mark.
